Abstract:通过PCR法从奥利亚罗非鱼DNA中扩增出MSTN基因。该序列全长1 957 bp,含有3个外显子,2个内含子。外显子Ⅰ379 bp,外显子Ⅱ371 bp,部分外显子Ⅲ145 bp,内含子Ⅰ305 bp,内含子Ⅱ751 bp,编码区895 bp,共翻译298个氨基酸。通过随机测序法筛选SNPs,获得了6个突变位点,外显子Ⅰ、Ⅱ各有一个,而内含子Ⅱ则含有4个突变位点。外显子Ⅰ、Ⅱ的突变位点均导致翻译的氨基酸序列发生改变,为错义突变。研究结果为SNPs位点与奥利亚罗非鱼生长性能关联分析奠定了基础。

Abstract:从异育银鲫外周血中分离淋巴细胞,利用RPM I-1640培养基培养48 h后,采用流式细胞仪分析方法研究不同浓度的植物凝集素(phytohem agglutin in,PHA)(0μg/mL、80μg/mL、200μg/mL、400μg/mL、600μg/mL、800μg/mL、1 000μg/mL、1 200μg/mL)对外周血淋巴细胞转化率的影响,旨在探索异育银鲫外周血淋巴细胞体外转化最适刺激剂量。结果表明:(1)在PHA浓度0~400μg/mL的范围内,随着PHA浓度的增加,淋巴细胞转化率也增加,呈浓度依赖的关系,但当PHA的浓度增加到1 200μg/mL,淋巴细胞转化率明显下降。其中PHA浓度为400μg/mL时,淋巴细胞转化率最高,显著高于低浓度组(80μg/mL)和高浓度组(1000μg/mL、1200μg/mL)(P<0.05),略高于中浓度组(200μg/mL、600μg/mL、800μg/mL),差异不显著(P>0.05)。由此可见,在本试验条件下,PHA体外刺激异育银鲫淋巴细胞转化的最适剂量为400μg/mL。(2)用流式细胞仪检测细胞内DNA合成情况,Mu lticyc le AV软件进...

Abstract:通过室内60 d罗氏沼虾幼虾养殖试验,研究了不同养殖密度(150 ind/m3、400 ind/m3、550 ind/m3、800 ind/m3)对水质与幼虾生长特点的影响。试验发现,各密度组水质指标pH、DO、TAN、NH3-N、NO2--N和CODMn均控制在罗氏沼虾生长的适合范围之内,密度对主要水质指标未产生显著性影响;不同养殖密度对幼虾生长指标产生不同程度的影响,最低密度组成活率和增长率与其他各组存在显著性差异(P<0.05),次低密度组增长率与最高密度组存在显著性差异(P<0.05),最低密度组的增重率与其他各组存在极显著差异(P<0.01);4种密度组的罗氏沼虾幼虾体重(w)和体长(L)之间的关系都符合幂函数方程:w=aLb(a的范围为0.013 2~0.015 1,b的范围为3.433 0~3.502 1);体长(L)和养殖天数(d)间符合线性相关:L=ad+b(a的范围为0.038 5~0.049 2,b的范围为1.801 1~1.886 1)。室内高密度养殖罗氏沼虾幼虾可据自身养殖条件,参考150~400 ind/m3选用布苗密度。

Abstract:实验室条件下,研究了N、P浓度、不同化合态N及N∶P比值对坛紫菜(Porphyra haitanensis)N、P吸收速率,以及P浓度对坛紫菜生长速率和藻红蛋白含量的影响。结果表明,随着营养盐浓度的升高,坛紫菜对N、P的吸收速率也随之增高,当无机氮浓度达到100μmol/L时,坛紫菜对N、P的吸收速率趋向接近最大值;当NO3--N∶NH4+-N比值为1∶5时,坛紫菜对N的吸收达到最大值;坛紫菜对P的吸收速率随NO3--N∶NH4+-N比值的减小而略有增大;坛紫菜对N的吸收速率随着N∶P比值的增大而增大,而对P的吸收速率随着N∶P比值的增大而减小;在磷浓度低于12μmol/L的情况下,坛紫菜的生长速率和藻红蛋白含量随着P浓度的升高而增加,高于12μmol/L时,则不再增加。

Abstract:配制添加不同浓度壳聚糖(0、0.2%、0.4%、0.6%、0.8%、1.0%)的6组实验饲料,网箱养殖淡水白鲳(Colossoma brachypomum)(82.26±5.13)g 60 d,探讨壳聚糖对淡水白鲳生长、非特异性免疫功能的影响和其在饲料中的适宜添加量。结果表明:0.6%组末体重、特定生长率、饲料效率和蛋白质效率显著高于0、0.2%、0.4%和1.0%组(P<0.05);0.6%、0.8%和1.0%组脏体比显著低于0、0.2%和0.4%组(P<0.05);0.6%、0.8%和1.0%组肝体比显著低于0组(P<0.05);0.6%组溶菌酶活性最高,显著高于0和0.2%组(P<0.05);0.6%组超氧化物歧化酶活性最高,显著高于其它组(P<0.05);0.6%组酸性磷酸酶活性显著高于0和0.2%组(P<0.05);用嗜水气单胞菌对淡水白鲳进行攻毒后的14 d内,0.6%、0.8%和1.0%组的死亡率显著低于0组(P<0.05)。综合各项实验指标,淡水白鲳饲料中添加适量的壳聚糖能提高其生长性能、饲料利用率、非特异性免疫功能以及抗嗜水气单胞菌感染能力,适宜添加量0.6%~0.8%。

Abstract:研究了浒苔(Enteromorpha prolifera)和条浒苔(Enteromorpha clathrata)在室内条件下的生长及其氨氮吸收动力学特征。结果表明:浒苔和条浒苔藻体体重日生长率分别为17.30%和16.82%;浒苔和条浒苔藻体幼苗在温度25℃和光照140μmol/(m2.s)的条件下达到最大体长日生长率,分别为78.9%和82.1%。在1~10 g/L密度范围内,浒苔和条浒苔对NH4+-N的吸收速率随密度和时间的增加而增加,当藻体密度为10 g/L时,NH4+-N浓度分别下降了86.43%和84.13%。两种浒苔对NH4+-N吸收速率与介质中NH4+-N浓度呈显著的线性关系,在NH4+-N浓度为400μmol/L时,30 min后浒苔和条浒苔的吸收速率分别为421和409μmol/(gDW.h),说明两种浒苔吸收氨氮的方式以被动扩散为主。在400μmol/L的起始浓度下,浒苔和条浒苔对NH4+-N的吸收速率随时间变化呈现3个不同阶段:在75 min内呈快速吸收阶段,75~185 min为内部NH4+-N控制的吸收阶段,250 min后为外界NH4+-N浓度控制的吸收阶段;在700 min后,...
